Peptide purity testing distinguishes several impurity classes. Related substances include truncated sequences, deletion peptides, and diastereomers formed during synthesis, while residual solvents, counterions, and water are not peptide-related but affect mass balance. Aggregates and oxidation products can arise during storage. Each class requires different analytical approaches, and a complete purity profile combines separation, mass measurement, and orthogonal assays. Reporting only a single percentage can obscure which impurities are present, so the profile should name the methods and limits used.
Quality control relies on predefined specifications rather than a single purity number. A certificate of analysis typically lists the test method, acceptance limit, and measured result for each attribute. Common specifications include appearance, peptide content, water content, counterion identity, and related substances. Limits are set according to the peptide's intended use and the capability of the analytical method. A result outside a limit triggers investigation, not automatic rejection, because method variability and sample handling can affect outcomes.
Sample handling influences measured purity. Lyophilized peptides are hygroscopic and can absorb water, changing weight-based calculations, while repeated freeze-thaw cycles may promote aggregation or degradation. Dissolved samples should be prepared fresh when possible and protected from light and heat. In purity testing, the same handling conditions should apply to standards and samples. Stability-indicating methods are designed to separate degradation products from the parent peptide, though open questions remain about how accelerated stability data predict long-term behavior for every sequence.
Stability testing examines how purity changes under controlled conditions. Samples are stored at defined temperatures, such as -20 °C or -80 °C, and analyzed at intervals. Lyophilized powders are generally more stable than solutions because water promotes hydrolysis and aggregation. Repeated freeze-thaw cycles can also degrade peptides, especially those with oxidation-prone residues. Accelerated studies at elevated temperature provide useful comparisons, but they do not always predict long-term behavior at lower temperatures.
Handling practices influence measured purity. Peptides may adsorb to plastic or glass surfaces, particularly when hydrophobic or positively charged. Weighing hygroscopic powders can introduce water and alter concentration. Dissolving in appropriate solvents and using low-binding tubes can reduce losses. Each laboratory should validate its own procedures because recovery and stability vary with peptide sequence, formulation, and container material. Open questions remain about how best to standardize stability reporting across different peptide classes.

Orthogonal methods reduce the chance that a single technique misses an impurity. Capillary electrophoresis separates by charge-to-size ratio and can resolve variants that co-elute under one set of HPLC conditions. Amino acid analysis reports composition after hydrolysis and confirms the presence of expected residues. Karl Fischer titration measures water content, while ion chromatography can quantify counterions. No single number captures all aspects of sample quality, so reports often combine several measurements.
Peptide purity testing uses separation methods to estimate the proportion of a sample that corresponds to the target sequence. Reverse-phase high-performance liquid chromatography is the most common technique, separating peptides by hydrophobicity on a nonpolar column. Ultraviolet detection at 214 nm records peptide bonds and aromatic residues. The resulting chromatogram is reported as area percent, which reflects relative absorbance rather than absolute mass. This distinction matters because water, counterions, and residual solvents do not appear in the peptide peak.
Mass spectrometry provides an identity check that complements chromatographic purity. Electrospray ionization or matrix-assisted laser desorption/ionization measures the mass-to-charge ratio of intact peptides. A match to the expected molecular mass supports correct sequence length and terminal groups. Mass accuracy alone does not prove that every peak in a liquid chromatogram is the target peptide. It also does not directly quantify how much water or counterion remains in a lyophilized powder.
Handling and storage influence measured purity, and peptides can oxidize, deamidate, aggregate, or adsorb to surfaces over time. Lyophilized powders stored at -20 °C or lower are generally more stable than solutions, though some sequences require different conditions. Repeated freeze-thaw cycles can promote aggregation and loss, so testing after storage checks whether purity has changed. Stability-indicating methods compare stressed and unstressed samples to detect degradation pathways. Light exposure and pH can also accelerate modification.
Solid-phase peptide synthesis can produce truncated sequences when coupling reactions fail. Deletion peptides lack one or more internal residues, while truncation peptides end prematurely. Side reactions include aspartimide formation, oxidation of methionine, and aggregation during chain assembly. Crude synthetic peptides therefore contain target peptide plus related impurities, counterions, residual solvents, and water. Purification by preparative chromatography reduces these impurities but does not remove every closely related species, including some that differ by a single amino acid.

The Unison trade union suspends further school strikes in Scotland after receiving a revised pay offer from the Scottish Government, which will be put to its members in a ballot. Fiona, a sheep believed to have been stranded on the banks of the Moray Firth for two years, is rescued. 4 November – Pro-Palestinian protestors occupy two of Scotland's major railway stations, Edinburgh Waverley and Glasgow Central, during another day of protests as the war in Gaza continues. 5 November – Eight police officers are injured following Bonfire Night clashes with youths during which fireworks and petrol bombs are thrown; the most serious incident takes place in the Niddrie area of Edinburgh, where 50 youths clash with police. Two people are subsequently arrested over the Niddrie incident. Humza Yousaf's parents in-laws return to Scotland.
For copper, the CTAP-1 sensor shows a response in the UV region when Cu(I) binds to an azatetrathiacrown motif that in turn excites a pyrazoline-based dye that is attached. In Coppersensor-1 (CS1), a thioether-rich motif binds to Cu(I) causing the excitation of a boron-dipyrromethene (BODIPY) dye in the visible region. Iron sensors include Pryrene-TEMPO, in which the binding of iron to TEMPO quenches the fluorescence of pyrene when no Fe(II) is bound. Upon binding however, TEMPO is reduced and pyrene regains fluorescence. This probe is limited in that an analogous response can be generated by unwanted free radicals, and that it can only by used in acidic solution. The DansSQ Fe(II)-binding system consists of a Dansyl group bound to styrylquinoline and operates by the disruption of intra-molecular charge transfer. It is limited in that it is only soluble in acetonitrile in 10% H2O. Cobalt sensors have been made that capitalize on the breaking of C-O bonds by Co(II) in a fluorescent probe known as Cobalt Probe 1 (CP1).
Salyut 1, the first space station, was launched by the Soviets on April 19, 1971. Three days later, the Soyuz 10 crew attempted to dock with it, but failed to achieve a secure enough connection to safely enter the station. The Soyuz 11 crew of Vladislav Volkov, Georgi Dobrovolski and Viktor Patsayev successfully docked on June 7, and completed a then record 22-day stay. The crew became the second in-flight space fatality during their reentry on June 30, when they were asphyxiated due to the spacecraft's cabin becoming depressurized, shortly after undocking. The disaster was blamed on a faulty cabin pressure valve, that allowed the air to vent into space. The crew was not wearing pressure suits and had no chance of survival once the leak occurred. To prevent a recurrence of the Soyuz 11 tragedy, Soviet engineers redesigned the Soyuz spacecraft and mandated that cosmonauts wear Sokol pressure suits during launch and landing, a requirement still in place today. The United States launched a single orbital workstation, Skylab, on May 14, 1973. It was launched using a leftover Saturn-5 rocket from the Apollo programme. Skylab weighed 169,950 pounds (77,090 kg), was 58 feet (18 m) long by 21.7 feet (6.6 m) in diameter, and had a habitable volume of over 10,000 cubic feet (280 m3). Skylab was damaged during the ascent to orbit, losing one of its solar panels and a meteoroid thermal shield. Subsequent crewed missions repaired the station, and conducted valuable research.

== Occurrence as natural products == There is a widespread occurrence of the 2,5-diketopiperazine core in biologically active natural products. The most structurally diverse 2,5-diketopiperazine natural products are based on tryptophan and proline modified by heterocyclisation and isoprenyl addition. These range from the hepatoxic brevianamide F (cyclo(L-Trp-L-Pro)) to the annulated tremorogenic verruculogen and the spiro-annulated spirotryprostatin B which represent a promising class of antimitotic arrest agents, to the structurally complex (+)-stephacidin A, a bridged 2,5-diketopiperazine that possess a unique bicyclo[2.2.2]diazaoctane core ring system and is active against the human colon HCT-116 cell line.

Estrogen sulfates like estradiol sulfate or estrone sulfate are about twice as potent as the corresponding free estrogens in terms of estrogenic effect when given orally to rodents. This in part led to the introduction of conjugated estrogens (Premarin), which are primarily estrone sulfate, in 1941. Although inactive at steroid hormone receptors, E2S has been found to act as a potent inhibitor of glutathione S-transferase, an enzyme that contributes to the inactivation of estradiol via conversion of it into an estradiol-glutathione conjugate. As such, E2S can indirectly serve as a positive effector of estrogen signaling. Estradiol levels are about 1.5- to 4-fold higher than E2S levels in women. This is in contrast to E1S, the levels of which are about 10 to 15 times higher than those of estrone. E2S at an oral dosage of 5 mg/day in women resulted in inhibition of ovulation in 89% of cycles (47 of 53).
== Angolan front (1975–1977) == On 25 April 1974, the Carnation Revolution ousted Marcelo Caetano and Portugal's right-wing Estado Novo government, sounding the death knell for the Portuguese Empire. The Carnation Revolution was followed by a period of instability in Angola, which threatened to erupt into civil war, and South Africa was forced to consider the unpalatable likelihood that a Soviet-backed regime there allied with SWAPO would in turn create increased military pressure on South West Africa. PLAN incursions from Angola were already beginning to spike due to the cessation of patrols and active operations there by the Portuguese. In the last months of 1974, Portugal announced its intention to grant Angola independence and embarked on a series of hasty efforts to negotiate a power-sharing accord, the Alvor Agreement, between rival Angolan nationalists. There were three disparate nationalist movements then active in Angola, the People's Movement for the Liberation of Angola (MPLA), the National Union for the Total Independence of Angola (UNITA), and the National Liberation Front of Angola (FNLA). The three movements had all participated in the Angolan War of Independence and shared a common goal of liberating the country from colonial rule, but also claimed unique ethnic support bases, different ideological inclinations, and their own conflicting ties to foreign parties and governments. Although each possessed vaguely socialist leanings, the MPLA was the only party which enjoyed close ties to the Soviet Union and was openly committed to Marxist policies.

monoclonal Describing cells, proteins, or molecules descended or derived from a single clone (i.e. from the same genome or genetic lineage) or made in response to a single unique compound. Monoclonal antibodies are raised against only one antigen or can only recognize one unique epitope on the same antigen. Similarly, the cells of some tissues and neoplasms may be described as monoclonal if they are all the asexual progeny of one original parent cell. Contrast polyclonal.
==== France ==== On 5 June 1916, Tolkien boarded a troop transport for an overnight voyage to Calais. Like other soldiers arriving for the first time, he was sent to the British Expeditionary Force's base depot at Étaples. On 7 June, he was informed that he had been assigned as a signals officer to the 11th (Service) Battalion, Lancashire Fusiliers. The battalion was part of the 74th Brigade, 25th Division. While waiting to be summoned to his unit, Tolkien sank into boredom. To pass the time, he composed a poem titled The Lonely Isle, which was inspired by his feelings during the sea crossing to Calais. To evade the British Army's postal censorship, he developed a code of dots by which Edith could track his movements. He left Étaples on 27 June 1916 and joined his battalion at Rubempré, near Amiens. He found himself commanding enlisted men who were drawn mainly from the mining, milling, and weaving towns of Lancashire. According to John Garth, he "felt an affinity for these working class men", but military protocol prohibited friendships with "other ranks". Instead, he was required to "take charge of them, discipline them, train them, and probably censor their letters ... If possible, he was supposed to inspire their love and loyalty." Tolkien later lamented, "The most improper job of any man ... is bossing other men. Not one in a million is fit for it, and least of all those who seek the opportunity."
Consecutive treaties between the Ottoman Empire and the Polish–Lithuanian Commonwealth called for the governments to keep the Cossacks and Tatars in check, but neither enforced the treaties strongly. The Polish forced the Cossacks to burn their boats and stop raiding by sea, but the activity did not cease entirely. During this time, the Habsburg monarchy sometimes covertly hired Cossack raiders against the Ottomans, to ease pressure on their own borders. Many Cossacks and Tatars developed longstanding enmity due to the losses of their raids. The ensuing chaos and cycles of retaliation often turned the entire southeastern Polish–Lithuanian Commonwealth border into a low-intensity war zone. It catalyzed escalation of Commonwealth–Ottoman warfare, from the Moldavian Magnate Wars (1593–1617) to the Battle of Cecora (1620), and campaigns in the Polish–Ottoman War of 1633–1634.
== History == In Ireland, during the Famine, prior to 1848, brown bread was handed out to the poor. In England, brown bread was made from brown meal. Around and prior to the year 1845, brown meal was considered a less desirable grain product, and was priced accordingly. However, by 1865, due to recently discovered health benefits of bran, brown meal's London price had increased to a point often greater than that of fine flour.
== As a drug target == With its integral role in HIV replication, HIV protease has been a prime target for drug therapy. HIV protease inhibitors work by specifically binding to the active site by mimicking the tetrahedral intermediate of its substrate and essentially becoming “stuck,” in the catalytic cycle, disabling the enzyme. After assembly and budding, viral particles lacking active protease cannot mature into infectious virions. Several protease inhibitors have been licensed for HIV therapy. There are ten HIV-1 PR inhibitors that are currently approved by the Food and Drug Administration: indinavir, saquinavir, ritonavir, nelfinavir, lopinavir, amprenavir, fosamprenevir, atazanavir, tipranavir, and darunavir. Many of the inhibitors have different molecular components and thus different mechanistic actions, such as blocking the active site. Their functional roles also extend to influencing circulation concentrations of other inhibitor drugs (ritonavir) and being used only for certain circumstances in which the virus exhibits tolerance of other inhibitors (tipranavir).

A related substance is a peptide-like impurity that resembles the target sequence, such as a truncated or modified form. It is often reported as individual and total area percent.
Each test measures a different attribute, such as identity, purity, water content, or counterion. Together they give a more complete quality profile than a single purity percentage.
Lyophilized powder is typically stored cold and dry, while solutions are kept cold and used promptly. Repeated freeze-thaw cycles should be avoided because they can change the sample.
It typically includes the peptide sequence, molecular mass, purity method and result, storage recommendations, and date of analysis. Raw chromatograms and mass spectra may be provided on request. The absence of method details makes a purity value difficult to interpret.
