== Adverse effects == There is a need for controlled clinical trials before the safety of ibogaine can be established; existing evidence, largely from uncontrolled case reports, links it to significant acute and long-term adverse events (especially cardiac issues like QTc prolongation), including fatalities. It should only be used in medically supervised settings with thorough screening and continuous cardiac and psychiatric monitoring. Ibogaine can cause various cardiovascular changes (e.g., slowed heart rate, blood pressure shifts), neurological effects (ataxia, tremor, mania), and rare but serious complications, including arrhythmias, respiratory failure, and death, especially when combined with opioids or in patients with underlying conditions. Cardiac risks likely occur through hERG channel blockade leading to QT prolongation and arrhythmias; the synthetic analog 18-MC appears to produce much less of this dangerous effect despite similar binding affinity. A 2022 review found 58 ibogaine-associated emergencies, including 38 deaths. Two of these deaths occurred in the setting of clinical studies. According to a 2012 estimate, 1 in every 300 patients who take ibogaine die.
=== Sialorrhea === Sialorrhea is a condition where oral secretions are unable to be eliminated, causing pooling of saliva in the mouth. This condition can be caused by various neurological syndromes such as Bell's palsy, intellectual disability, and cerebral palsy. Injection of botulinum toxin type-A into salivary glands is useful in reducing the secretions.

A Cochrane review found suvorexant to be effective in the short-term treatment of sleep disturbances in people with dementia with few adverse effects. It is unknown if suvorexant is effective and safe for treatment of sleep problems in children and adolescents as suvorexant has not been studied in this context.

Both the Sammādiṭṭhisutta and the Mahānidānasutta specifically mention the factors of dependent origination as being related to the process of conception in the womb. Bhikkhu Bodhi affirms the centrality of rebirth for dependent origination. Bodhi writes that "the primary purpose, as seen in the most archaic Buddhist texts, is to show the causal origination of suffering, which is sustained precisely by our bondage to rebirth."
Clonidine may be used to ease drug withdrawal symptoms associated with abruptly stopping the long-term use of opioids, alcohol, benzodiazepines, and nicotine. It can alleviate opioid withdrawal symptoms by reducing the sympathetic nervous system response such as tachycardia and hypertension, hyperhidrosis (excessive sweating), hot and cold flashes, and akathisia. It may also be helpful in aiding smokers to quit. The sedation effect can also be useful. Clonidine may also reduce severity of neonatal abstinence syndrome in infants born to mothers that are using certain drugs, particularly opioids. In infants with neonatal withdrawal syndrome, clonidine may improve the neonatal intensive care unit Network Neurobehavioral Score. Clonidine has also been suggested as a treatment for rare instances of dexmedetomidine withdrawal.

=== Antisense pharmaceuticals === In the United States, the Food and Drug Administration (FDA) has approved the phosphorothioate antisense oligonucleotides fomivirsen (Vitravene) and mipomersen (Kynamro) for human therapeutic use in antisense therapy. To prevent degradation of the therapeutic oliogoneucleotides, chemical modification is usually required. The most common chemical modification on the oligonucleotides is adding a phosphorothioate linkage to the backbones. However, the phosphrothioate modification can be proinflammatory. Adverse effects including fever, chills or nausea have been observed after local injection of phosphrothioate modified oligonucleotides. Gapmers often utilize nucleotides modified with phosphorothioate (PS) groups. Miravirsen is an antisense phosphorothioate oligonucleotides that is additionally a locked nucleic acid gapmer. Other antisense oligonucleotides using phosphorothioates include afovirsen, aganirsen, alicaforsen, bepirovirsen, custirsen, drisapersen, eplontersen, evazarsen, gataparsen, inotersen, IONIS-GCCRRx, nusinersen, oblimersen, olezarsen, pelacarsen, sefaxersen, tofersen, and volanesorsen.
